superfolder GFP with N-terminal pdu-microcompartment tag

We have used superfolded GFP with the N-terminal pdu-microcompartment tag in order to get fluorescence images that would show localised spots of higher GFP-density, indicating that the microcompartment tag successfully inserts the attached protein into the microcompartments

To test the targeting of the N-terminal tag, we have used Fluorescence microscopy on cells that have been transformed with two vectors, one containing this part and one containing the microcompartment submitted by the iGEM Dundee team in 2009 ( BBa_K562009 ). As can be seen for some of the cells in the image below, there is no perfect and uniform distribution of sfGFP, but clearly identifiable spots of higher fluorescence densities which suggests that the targeting mechanism of the N-terminal tag has worked successfully.
